Either talk to your developer, or follow the instructions at Introduction for site developers in order to get a copy of the site that you can work on.

Manual approach

Find the list of glossary entries in: _data/glossary.csv
Edit or add entries as required - making sure that the definitions are surrounded by quotes

The first time a glossary entry appears on a page (outside of a heading), it will be highlighted with a link. Users can click on it to open the definition.
